The details depend on what power you need. That is 500 volts at how many milliamps at the output? The basic design that will suit most cases is a push-pull transformer inverter. It can be up to 30W. The first link below (geocities) shows a suitable version which is intended to work with the flyback transformer from a TV set to get very high voltages. However you can take all the wires off and just use the core to wind your own transformer. The turns that connect to the transistor are as stated. The 500V winding would be around 250 turns of enamelled wire. Make sure there is plenty of quality insulation between the high voltage and the other windings. If the transistor's windings are on the outside, they can be easily replaced if the voltage is wrong. Just adjust the number of turns to correct the turns ratio error. The wire should be 0.8 mm or more in diameter. The high voltage winding can be 0.1mm or whatever you can get. I think this supply could deliver 20W. It needs a bridge rectifier and high voltage reservoir capacitor added for completeness. This supply has enough power to kill you. Better not to play with such things if you don't have appropriate skills.

If the power is lower, use smaller transformer and transistors. Small mains transformers can also be used for this circuit by connecting them in reverse. They are not all that efficient. The second link below(members) shows how to do that. The second link on that page (10-15w watt 100% Radio Shack DC-DC Converter) is the one to use.
Note that it uses a voltage doubling rectifier. A 240V transformer with two 9V windings and a voltage doubler rectifier should do it. It can be rated about twice the power level you want. At powers below about 1W the 555 timers could drive the transformer directly without the transistors.
